A Comprehensive Review on the Health Benefits of Celery Seed Extract: Highlighting the Importance of Phthalides (Standardized to 3-N Butyl Phthalide and Sedanolide)

Celery seed extract, derived from the seeds of Apium graveolens, has been used in traditional medicine for its numerous health benefits. It is rich in phytochemicals, such as flavonoids, coumarins, and volatile oils, which contribute to its therapeutic properties. Historically, celery seed has been employed to treat conditions like gout, arthritis, and various inflammatory disorders. Recently, it has gained attention for its potential cardiovascular and neuroprotective benefits, primarily due to the presence of bioactive compounds such as Phthalides including 3 n-butylphthalide (3nBP) & Sedanolide.

Benefits of Unique Bioactive Profile

Celery seed extract contains several bioactive compounds, the most notable being Phthalide including 3-n-Butylphthalide (3nB) & Sedanolide, which are believed to be responsible for its therapeutic effects. These compounds have been shown to exert significant antioxidant, anti-inflammatory, diuretic, neuroprotective, and bone health-promoting properties.

The identified bioactive profile of celery seed standardized to Phthalides (3-N Butylphthalide & Sedanolide) has been extensively studied and researched to improve the conditions of:

  • Hypertension or High Blood Pressure: A common condition which can deteriorate the cardiovascular health by being a significant risk factor for fatal and non-fatal cardiovascular events. Studies have shown that proper blood circulation reduces the risk of Hypertension and its associated effects such as Stroke, myocardial infarction and other cardiovascular events.[1]
  • Hyperuricemia: An arthritic condition which can reduce the quality of life due to the pain, swelling, redness at the affected joints. The condition of Hyperuricemia, or commonly called Gout, is observed when the body produces more Uric acid than normal range (normal range of uric acid is up to 7 mg/dl)[2] or when Uric acid is not excreted out properly through Kidney. Both conditions lead to the deposition of monosodium urate (MSU) crystals or gout stones at joints causing severe pain, inflammation and redness.

Reducing the production of Uric acid along with proper excretion of MSU can improve the condition of Gout. Also, lowering inflammatory markers in the joints and supporting proper kidney function, can additionally help in improving the condition of Gout.

Celery Seed Extract Standardized to Total primarily 3-N Butyl Phthalide and Sedanolide, can improve the quality of life by improving the condition of Hypertension and Joint Health.

Rationale for Celery seed Extract in Hypertension

The unique bioactive profile standardized to Total Phthalides including 3-NBP and sedanolide has been well studied in various Preclinical and Clinical Studies to understand its mechanistic approach and its therapeutic potential in treatment of Hypertension.

One such study discussed the vasodilatory effects of Celery Seed extract on endothelial tissues. A preclinical in-vitro and in-vivo study explains the vasorelaxation of endothelial cells by the following mechanism of action:

Inhibition of Calcium/Ca2+ channel: Vasoconstriction of the endothelial cells especially aortic smooth muscle cells becomes high when there is influx of extracellular Calcium/Ca2+ ions. Studies suggest Butyl Phthalides has the same mechanistic approach as calcium channel blockers thus reducing the Phosphorylation of Myosin heads of Smooth muscle cells. This reduced phosphorylation causes vasorelaxation. The calcium blockade property of Butyl Phthalides is assumed to be responsible for showing Hypotensive effects.[3]

Human Clinical Data: Improving the Systolic/Diastolic Blood Pressure

In 30 Mild to Moderate Hypertensive Patients, it was found that a standardized celery seed extract, rich in Phthalides including 3-N Butyl Phthalate has shown clinically relevant blood pressure-lowering effects. This study evaluated the potential of standardized Celery seed extract in showing the hypotensive property. After the 6 Week Study, it was found that there is a significant decrease in both systolic blood pressure (SBP) and diastolic blood pressure (DBP) compared to baseline.

The authors of the study concluded that significant reduction in SBP & DBP is due to vasodilatory property of 3-NBP mediated by its calcium channel inhibiting property. They further concluded that the findings of the study suggest celery seed extract can be potential

alternative to present day medications for hypertension as celery seed extract does not inhibit Angiotensin Converting Enzyme (ACE) which lowers the cerebral blood flow. [4]

Rationale for Celery Seed Extract in Hyperuricemia

Role of Flavonoids & Phthalides

Flavonoids and Phthalides constitute the major active compounds in celery seeds. These compounds have been extensively studied through various preclinical and clinical studies for their therapeutic potential. The studies suggest their anti-inflammatory property, diuretic property and antioxidant properties can be an alternative to various conventional medicines.

One such study highlights the potential of this unique composition of flavonoids and Phthalides as an alternative for improving the condition of Chronic Hyperuricemia. Studies suggest mechanistic pathways that a standardized celery seed extract follows to reduce chronic conditions of Hyperuricemia:

  • Xanthine Oxidase Inhibitor: Purines from foods and body cells follow a pathway where the enzyme, xanthine oxidase, acts on intermediates hypoxanthine and xanthine to form uric acid. The bioactive of the celery seed extract acts as xanthine oxidase inhibitor and thus prevents the enzymatic action required for conversion of purines to uric acid.[1]A few research also indicates that flavonoids and phthalides in the celery seed extract show diuretic property which improves the excretion of uric acid in natural as well as crystal forms.[5]

A broad spectrum of systematic reviews and scientific data demonstrated that fenugreek and its extracts improve glycemic and lipid profiles. Fenugreek is known to have antioxidant, anti-carcinogenic, gastro protective, anti-inflammatory, antimicrobial, immunological, anti-obesity, and hepatoprotective effects, and are beneficial to women’s health. Further, previous studies reported that fenugreek improved cognitive functions and Parkinson’s symptoms and showed anti anxiety, antidepressant, and neuroprotective properties. A recent study has reported the beneficial effects of fenugreek and its extract on hormonal-related statuses, such as galactagogue in lactating women and male impotence.

The presence of fenugreek seeds in various traditional medicinal systems is due to their composition of diverse phytochemicals, including Glycosides.

Potential benefits of fenugreek glycosides in testosterone production include: Increased Testosterone Levels:

  • Fenugreek supplementation may lead to an increase in testosterone levels, which could have positive effects on muscle mass, strength, and overall vitality.
  • Enhanced Libido: Fenugreek has been traditionally used as an aphrodisiac, and its potential influence on testosterone levels may contribute to improvements in libido and sexual health.
  • Improved Exercise Performance: Higher testosterone levels may be associated with increased exercise performance and muscle strength, making fenugreek an area of interest for athletes and those involved in physical fitness.
  • Spermatogenesis Support: Testosterone plays a crucial role in spermatogenesis (sperm production), and fenugreek’s potential impact on testosterone levels may have implications for male reproductive health.

Testosurge® is standardized to include a specific combination of steroidal and flavonoid glycosides. The steroidal glycosides in Testosurge® enhance the effectiveness of testosterone by competitively binding with SHBG. When testosterone is released into the bloodstream approximately 60-70% of it binds to SHBG. The steroidal glycosides from Testosurge® binds to SHBG and make testosterone free and bioavailable.

On the other hand, the flavonoid glycosides in Testosurge® play a role in increasing testosterone production. They achieve this by enhancing the expression of the steroidogenic acute regulatory (StAR) protein in Leydig cells. Testosterone is produced within the mitochondria of Leydig cells through steroidogenesis, where the StAR protein facilitates the entry of cholesterol into the mitochondrion.

Testosurge® has demonstrated inhibitory activity against5-alpha reductase and aromatase in cell line studies. 5-alpha reductase is an enzyme responsible for converting testosterone into DHT, while aromatase converts testosterone into Estrogen. This inhibition activity suggests that Testosurge® may help regulate the levels of these hormones in the body.

Additionally, Testosurge® can inhibit PDE5 enzyme which canresult in improved vasodilation and penile blood flow.
